Roads Minister Danny Kennedy announced the start of a £150,000 resurfacing scheme on Hillsborough Road at Hillsborough.
The work begins this week between Ballykeel Crossroads and Dromara.
Mr Kennedy said: "This scheme will increase the life of the road, greatly improving the strength and ride quality of the carriageway on an important route between Dromara and Hillsborough."
To ensure the safety of road users and the operation on site a full closure will be in place from Dromore Road, Hillsborough to Ballynahinch Road, Dromara.
Until 30 November, when the work is expected to be completed, works on the Hillsborough Road will commence each morning from approximately 7.30am and continue until 6.00pm each evening. During these works the carriageway will be down to single file traffic with signals in operation.
Closures will be required for the surfacing and during these periods all traffic will be diverted onto the B2 Ballynahinch Road and B7 Dromore Road.
